Description:
Myeloperoxidase(MPO)isaperoxidaseenzyme(EC1.11.1.7)mostabundantlyexpressedinneutrophilgranulocytes.Itisalysosomalproteinstoredintheazurophilicgranulesoftheneutrophil.MPOcontainsahemepigmentwhichcausesitsgreencolorinsecretionsrichinneutrophils,suchaspusandsomeformsofmucus.MPOcatalyzestheproductionofhypochlorousacid(HClO)fromhydrogenperoxide(H₂O₂)andchlorideanion(Cl-,ortheequivalentfromanon-chlorinehalide).MPOalsooxidizestyrosinetoatyrosylrADIcalusinghydrogenperoxideasanoxidizingagent.InHansABIoMed"sMPOAssayKit,theHClOproducedfromH₂O₂andCl-isreactedwithtaurinetogeneratethetaurinechloramine,whichsubsequentlyreactswiththeTNB2-probetoeliminatecolor(λ=412nm).Thekitprovidesarapid,simple,sensitive,andreliabletestsuitableforhighthroughputactivityassayofMPO.ThiskitcanbeusedtodetectMPOaslowas0.05mUperwell.
