Description:

GlutathionePeroxidase(GPx,EC1.11.1.9)familyofenzymesplayimportantrolesintheprotectionoforganismsfromoxidativedamage.GPxconvertsreducedglutathione(GSH)tooxidizedglutathione(GSSG)whilereducinglipidhydroperoxidestotheircorrespondingalcoholsorfreehydrogenperoxidetowater.Severalisozymeshavebeenfoundindifferentcellularlocationsandwithdifferentsubstratespecificity.LowlevelsofGPxhavebeencorrelatedwithfreerADIcalrelateddisorders.IntheGlutathionePeroxidaseActivityAssay,GPxreducesCumeneHydroperoxidewhileoxidizingGSHtoGSSG.ThegeneratedGSSGisreducedtoGSHwithconsumptionofNADPHbyGR.ThedecreaseofNADPH(easilymeasuredat340nm)isproportionaltoGPxactivity.Theassaycanbeusedtomeasurealloftheglutathionedependentperoxidasesinplasma,erythrocytelysates,tissuehomogenates,andcelllysateswithadetectionsensitivityof~0.5mU/mlofGPxinsamples.
